How do I transfer an agar culture into liquid media?

Below are two easy ways to transfer an agar culture into liquid media. Please note that it is recommended to use a 1:5 ratio of algae to new media when scaling up to larger volumes:

  1. Use a sterile inoculating loop or sterile wooden stick, scrape the algae off of the agar slant. Then 'swish' the loop in sterile liquid media to inoculate it.
  2. Use a sterile pipette to put a few milliliters of sterile liquid media into the agar tube. Use the pipette tip to scrape the algae off of the agar and into the liquid media. Pipette the media+algae mixture back out of the agar tube, and transfer it into the new vessel containing liquid media.
